Yüksek hassasiyeti ile jel bazlı ya da kapiler elektroforez bazlı sistemlerin yakalayamadığı mutasyonları veya sekans sistemlerinin tespit edemediği somatik mutasyonları tespit edebilir. Kanser ile bağlantılı genlerin büyüklüğü ve mutasyon frekanslarının düşüklüğü nedeniyle kanser mutasyonu tanısı için yaygın olarak kullanılmaktadır. Sekanslama işlemi öncesi DHPLC sistemiyle yapılan ön mutasyon taraması, maliyet ve zamandan ciddi ölçüde tasarruf edilmesine olanak sağladığından DHPLC’nin dünya çapında ideal bir ’’sekans öncesi tarama’’ olarak kabül görmesini sağlamıştır. Dünya çapında kurulu yaklaşık 1200 sistemden günümüze kadar çeşitli alanlarda 1100’ün üzerinde makale yayınlanmıştır.
DHPLC Teknolojisi Mutasyon veya polimorfizm içeren tek zincir DNA molekülleri, yabanıl tip bir tek zincir DNA molekülü ile hibridize edildiğinde, heteroduplexler oluşur. Bu heteroduplekslerin erime sıcaklığı homoduplekslerden daha düşüktür. Kısmi eritilmiş heterodupleksler, yabanıl tip homoduplekslerden WAVE® sisteminde kolayca ayrılabilmektedir.
